In dairy cows, the current study investigated the applicability of a calving prediction model trained using supervised machine learning and ruminal temperature (RT) data. An investigation into cow subgroups experiencing prepartum RT changes included a comparison of the model's predictive performance across these subgroups. Real-time data from 24 Holstein cows were collected at 10-minute intervals using a real-time sensor system. Determining residual reaction times (rRT) involved calculating the average hourly reaction time (RT) and representing the data as deviations from the mean reaction time for the same hour over the previous three days (rRT = actual RT – mean RT for the same time on previous three days). Starting around 48 hours before the cow delivered her calf, the average rRT decreased consistently, reaching a minimum of -0.5°C five hours before calving. In contrast, two classifications of cows were observed: a first cluster (n = 9) marked by a late and modest rRT reduction, and a second cluster (n = 15) characterized by an early and substantial rRT decrease. A support vector machine was employed to develop a calving prediction model based on five features derived from sensor data, which characterize prepartum rRT changes. A novel, de novo mutation in SPTLC1 (c.58G>A, p.A20T) was found in a sporadic patient. In a group of 16 JALS patients, 7 carried FUS mutations, and 5 demonstrated mutations in SPTLC1, SETX, NEFH, DCTN1, and TARDBP. Patients with SPTLC1 mutations showed an earlier age of onset (7946 years) than patients with FUS mutations (18139 years) (P <0.001), accompanied by significantly prolonged disease duration (5120 [4167-6073] months) in contrast to FUS mutation patients (334 [216-451] months, P <0.001). For a better representation of the structure and function of airway smooth muscle in small airways, microtissues with toroidal ring shapes are exceptionally well-suited, leading to a deeper understanding of diseases like asthma. To create microtissues shaped as toroidal rings, polydimethylsiloxane devices that contain a series of circular channels surrounding central mandrels are used to facilitate the self-aggregation and self-assembly of airway smooth muscle cell (ASMC) suspensions. Gradually, the ASMCs in the rings transition to a spindle shape, then align axially along the ring's circumference. Over 14 days of culture, the strength and elastic modulus of the rings increased, while the ring size remained largely unchanged. Gene expression profiling indicated stable expression of messenger RNA molecules for extracellular matrix proteins, including collagen type I and laminins 1 and 4, maintained over a period of 21 days in cell culture. TGF-1's influence on cells within the rings leads to a notable decrease in ring circumference and a rise in the levels of extracellular matrix and contraction-related mRNA and protein.
